近日，Certa Therapeutics公布其靶向G蛋白偶联受体（GPCR）拮抗剂FT011的2期临床试验积极数据。结果显示，经过12周的治疗后，超过60%的硬皮病患者获得了具有临床意义的改善。
硬皮症（scleroderma）是一种以皮肤和其他器官（通常是肺、肾和心脏）发炎和纤维化为特征的自身免疫性疾病。患者通常会出现活动能力丧失、疼痛、疲劳，这将严重损害患者的生活质量并影响其心理健康。该病有可能会危及生命，其死亡率是风湿性疾病中最高的。
FT011是一种潜在“first-in-class”口服疗法，用于治疗多种器官的慢性纤维化。FT011是Certa开发的多个新型化合物之一，靶向先前尚未成药过的一个GPCR靶点。这种GPCR蛋白在健康组织中沉默，但在受伤或患病后会被激活。有证据显示，该GPCR在导致炎症和纤维化的多个下游通路中发挥作用。
